Actually, usually plasmids are cut in two places, using two restriction enzymes, and so a small section is usually cut out. I ‘lose’ about 10-50bp from a non-coding region of pJK11 when I digest it in preperation for ligation, depending on which enzymes I’m using.

We use enzymes produced naturally by living things. These enzyme only cut DNA where it forms a particular pattern. e.g. HindIII cuts the code AAGCTT.
The girl actually skips over many steps in the process (Primer design, PCR, gel electrophoesis, plasmid ligation etc). You would actually extract the DNA from human cells and perform most steps in vitro; the digestion/cutting would happen in a test tube (well, I use a microcentrifuge tube).

This ninth grader just taught me, a 20 year old, a lot more than my college professor ever did. Nice.

Religion was men for the middle age, and i don’t care what god you believe in, it’s imagination. Great video! Yes people out there who think that she has made this up, this is how they Make insulin for diabetics. So yes we are already using genetically modified organisms. If this this was at my school I’d give her a A++++
